Optimization of the formula for the compound ointment of Polygonatum sibiricum and lotus leaf and its modulation on hepatic lipid metabolism of high-fat diet rats(黄精荷叶复合膏配方优化及对高脂膳食大鼠肝脏脂质代谢的调节作用)

      Polygonatum sibiricum and lotus leaf were used as raw materials to produce a kind of compound ointment with lipid-lowering and liver-protecting activities. Using the sensory score, texture and water precipitation rate as indicators, the formula was optimized by single factor and orthogonal experiments. The pharmacological activities of compound ointment were evaluated in high-fat diet rats treated with 8 mg/kg and 16 mg/kg compound ointment by gavage administration. The results showed that the compound ointment of P. sibiricum and lotus leaf taste harmoniously with a thick and stable structure under the conditions of m (P. sibiricum ointment)∶m (lotus leaf ointment)=13∶7, sodium carboxymethylcellulose of 0.48%, oligoisomaltose of 48% and anhydrous citric acid of 6.0%, which is the best formula. The in vivo experimental results showed that the compound ointment significantly improved dyslipidemia, the accumulation of hepatic lipid and epididymal fat, and splenomegaly induced by the high-fat diet in rats (P<0.05). Furthermore, the liver function was investigated with results showing that the serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) enhanced by the high-fat diet could be significantly reversed by intervention with a high dose of compound ointment of P. sibiricum and lotus leaf (P<0.05). Moreover, a high dose of the compound ointment significantly reduced the mRNA levels of sterol-regulatory element binding protein-1c gene (SREBP-1c) and acetyl-CoA carboxylase α gene (ACC-α) and increased the mRNA level of carnitine palmitoyl transferase-1 gene (CPT-1). In conclusion, the edible quality of the compound ointment of P. sibiricum and lotus leaf was excellent, and it showed significant efficacy in protecting hepatic function and regulating hepatic lipid metabolism in high-fat diet rats. . 本研究以黄精、荷叶为原料,生产具有降脂护肝功效的复合膏。以感官评分、质构以及析水率作为指标,采用单因素及正交试验对复合膏的配方进行优化。通过高脂膳食喂养大鼠,分别灌胃给予8、16 mg/kg的复合膏液,探究其药理活性。结果显示:最佳的膏方配比为黄精膏与荷叶膏质量比为13∶7,羧甲基纤维素钠添加量为0.48%,低聚异麦芽糖添加量为48%,无水柠檬酸添加量为6.0%。配方优化后的黄精荷叶复合膏感官评分高,结构状态浓稠、稳定。体内实验结果表明,黄精荷叶复合膏可以显著改善高脂膳食诱导的大鼠血脂异常、附睾脂肪积累、肝脏脂质蓄积以及脾脏肿大(P<0.05)。进一步对大鼠肝功能进行探究,发现高脂膳食显著提高了血清中谷丙转氨酶(alanine aminotransferase, ALT)、谷草转氨酶(aspartate aminotransferase, AST)以及乳酸脱氢酶(lactate dehydrogenase, LDH)水平(P<0.05),而高剂量黄精荷叶复合膏干预可以显著逆转这些改变(P<0.05);此外,高剂量黄精荷叶复合膏显著降低了大鼠肝脏甘油三酯(triglyceride, TG)合成相关基因甾醇调节元件结合蛋白1c基因(sterol-regulatory element binding protein-1c gene, SREBP-1c)、乙酰辅酶A羧化酶α基因(acetyl-CoA carboxylase α gene, ACC-α)的mRNA表达量,提高了TG分解相关基因肉碱棕榈酰转移酶-1基因(carnitine palmitoyl transferase-1 gene, CPT-1)的mRNA表达量。综上所述,本研究制得的复合膏食用品质优良,在高脂膳食大鼠中表现出显著的保护肝功能、调节肝脏脂质代谢的功效。
