Rapeseed meal is an important protein source for animal nutrition especially for ruminants and monogastric animal species due to its well-balanced amino acid composition. There is also an increasing demand for vegetable proteins and the rapeseed protein could be used for human alimentation. However, high fiber content of the rapeseed meal limits protein content and availability in food and feed. The objective of this study was to analyze the inheritance of fiber related traits in four different F2 populations derived from crosses between five different genetic resources with lower lignin content in the seed. Parents and F2 populations were grown in the greenhouse of the Division of Crop Plant Genetics of the University of Göttingen, Germany. Fiber content was analyzed by wet chemistry analysis of defatted rapeseed meal by applying the ANKOM 200 filter bag. In addition, an NIRS calibration was used to predict fiber content. Two major QTLs linked to C03 and C05 genome were associated with reduced fiber content in rapeseed cultivars, thus a SNP based genotyping method- Kompetitive Allele Specific PCR (KASP) marker was used to observe allelic difference between parents and segregation ratio in F2 population. For this procedure, different markers associated to C03 and C05 were tested and polymorphic markers were used to observe allelic differences. Also, the genome of a candidate gene for lignin content – phenylalanine ammonia-lyase (PAL) gene was used to develop PCR-primers to study allelic sequence and observe differences among parental lines. Results from both PCR and KASP confirm differences between parents and segregation of lignin content in the F2-populations.
