      In this study, carbon nanomaterial-based disposable pencil graphite electrodes (PGE) were firstly developed and optimized. Accordingly, these electrodes were applied for the detection of interaction of activated protein C (APC) with its DNA aptamer. In this context, CNF-IL modified disposable graphite electrodes (CNF-IL/PGE) were prepared by modifying the surface of pencil graphite electrode with carbon nanofiber (CNF) and ionic liquid (IL) nanocomposite. The experiments for microscopic and electrochemical characterization of CNF-IL/PGE electrodes were then carried out. In this study, interaction between APC and its aptamer was performed in solution phase using APC-specific DNA aptamer (DNA-Apt), and electrochemical impedance spectroscopy (EIS) method was used in combination with CNF-IL/PGE in order to monitor the detection of biointeraction. The interaction of APC with its DNA aptamer as well as the interaction of APC with aptamer-antidote pair (AD22) were performed in solution phase and then impedimetric detection was performed by using CNF-IL/PGE. CNF-IL-PGE was used for the first time in the literature to detect impedimetrically the interaction of APC with its DNA aptamer, which will enable rapid, reliable and selective determination of APC in solution phase. The results obtained within the scope of the study will be a model for the development of new aptasensors that will provide the rapid and reliable sensitive and selective detection of similar biomolecular interactions. ; Çalışmamızda öncelikle karbon nanomalzeme temelli tek kullanımlık kalem grafit elektrotlar (PGE) geliştirilip ve optimize edilerek, ardından aktive edilmiş protein C (APC)'ye hedefli DNA aptameri ile APC'nin etkileşimine dayalı elektrokimyasal aptasensör geliştirildi.
