نبذة مختصرة : The pathogenic bacterium Streptococcus pneumoniae possesses two protein-kinases CpsD and StkP involved in polysaccharide capsule synthesis and cellular division respectively. Recently, a new bacterial protein-kinase family was identified, named UbK (Ubiquitous bacterial Kinase). My thesis work was to study the cellular role of UbK in the pneumococcus. I have demonstrated that Ubk is phosphorylated on a unique Tyrosine in vivo. A Δubk mutant, as well as catalytic (non ATPasic) or a permanently phosphorylated mutant shows growth and morphological defects. The deletion of ubk generates a suppressive mutation in the essential protein AsnS, whose role is assimilated to Asparaginyl-tRNA synthetase. Thus, I explored in details the link between UbK and AsnS. ubk is found in an operon induced in competence. I have evidenced that late competence and transformation are to a certain extent controlled by UbK. Morphological defects in absence of ubk appear to be typical of a cell-cycle impairment. We determined the cell wall composition was impaired, and that the UbK’s partners, FtsZ and FtsA that are implicated in cell division, are not correctly localized the Δubk mutant. Therefore, UbK is a major actor of cellular division and competence. ; La bactérie pathogène Streptococcus pneumoniae possède deux protéines-kinases CpsD et StkP impliquées respectivement dans la synthèse de la capsule polysaccharidique et de la division cellulaire.
