      Small cell lung cancer (SCLC) is highly aggressive with a poor prognosis (5-year survival rate of 7%). Paraneoplastic neurologic syndromes (PNS), characterized by autoimmune-mediated neuronal damage, occur in SCLC patients. The mechanistic link between SCLC and PNS remains unclear. This study investigates the role of the transcription factor Nuclear Factor 1 B (NFIB) in regulating Hu antigens, particularly in SCLC and PNS. We utilized data from mouse models and human SCLC cell lines to assess neuronal Hu antigen expression in association with NFIB levels. We observed significant upregulation of neuronal Hu antigens in NFIB-high SCLC cell lines. Mechanistically, NFIB directly regulated ELAVL2 expression via binding to its regulatory regions. Furthermore, neuronal Hu antigens exhibited subtype-specific expression patterns in neuroendocrine SCLC subtypes.
