Genotyp-Phänotyp-Untersuchung an einer Münchner Kohorte von Patienten mit krankheitsursächlichen Varianten in COL4A3 und COL4A4 assoziiert mit dem Alport-Syndrom und der Nephropathie vom Typ der dünnen Basalmembran ; Genotype-phenotype correlation in a munich cohort of patients with disease-causing variants in COL4A3 and COL4A4 associated with Alport syndrome and thin basement membrane nephropathy

Für eine Genotyp-Phänotyp-Untersuchung wurden von 89 Patienten, die mindestens eine als (wahrscheinlich) pathogen klassifizierte COL4A3-/COL4A4-Variante trugen, klinische Daten zu Prävalenz und Alter bei Erstmanifestation von mikroskopischer Hämaturie (MH), Proteinurie, terminaler Niereninsuffizienz und extrarenalen Manifestationen erhoben. Patienten mit einer monoallelischen nicht trunkierenden COL4A3-/COL4A4-Variante wiesen eine signifikant höhere Prävalenz von MH und Proteinurie auf als Patienten mit einer trunkierenden COL4A3-/COL4A4-Variante. ; For a genotype-phenotype correlation 89 patients carrying at least one COL4A3/COL4A4 variant classified as (likely) pathogenic according to the ACMG guidelines were recruited and clinical data concerning the prevalence and age of first manifestation of microscopic hematuria, proteinuria, end-stage kidney disease and extrarenal manifestations were raised. Patients with monoallelic non-truncating COL4A3/COL4A4 variants reported a significantly higher prevalence of MH and proteinuria than individuals with truncating COL4A3/COL4A4 variants.
